Stanley Kubrick. It would either be amazing or really, really boring.

That being said, there is a Dune movie, and from what I hear it's pretty good. Actually, I have it sitting around my hard drive, think I'll go watch it.

It is widely considered the pinnacle of Sci Fi writing. A lot of people like to make the comparison that Dune is to Sci Fi as Lord of The Rings is to Fantasy.

It would have to be like, David Cronenberg or Del Torro to direct, or even Riddley Scott, someone really out there as far as style.

As for casting, don't really matter. Sting played Leto Atriedes for god's sake. There ya go, Gene Simmons as Leto in Dune 2014

From what i've seen of it, Jodorowsky :P Otherwise, maybe Tarsem Singh, David Fincher, Alex Proyas or Park Chan Wook, ignoring whether or not they'd want to do it or if a studio would risk big money on any of them. They each have very different cinematic talents and a shared ability to create similarly dark, stylish and emotional films that juggle much more abrasive subjects than most directors, from violence through sexuality to sanity. If any of them wanted to do it and had a serious budget to work with they could smash it.

As for actors, possibly Chris Evans as Paul, Maisie Williams as Alia, Ian McShane as Leto, Idris Elba as Gurney, Gary Oldman as Duncan, Viggo Mortensen as Liet, Chiwetel Ejiofor as Stilgar, Emily Blunt as Chani, Tom Hardy as Feyd, as Baron Harkonnen and Ian McKellen as Shaddam. That's just a quick google-boosted crack at it, largely for fun, i can totally see other people in all these roles.

I should say that i'm a guy who loves what he knows of Dune, but i haven't read the books yet (i really want to). I've seen the film, which was a bit of a task, but an interesting one, and some of the mini-series, and i'm perfectly fine with adaptive changes being made where ever necessary/artistically worthwhile, so i'd be cool with changing ages and colours of characters for the sake of good actors playing them.

The sad truth is that Dune is a movie that can't, or more specifically shouldn't be made. The themes and concepts of politics, sex, religion, philosophy, spirituality and sociality run too deep in a book like Dune for it to be made accurately in to movie. In this day and age where everything needs to be a family friendly movie with a big budget that gets a big return there is no way a studio would put up the cash required to make something as weird and experimental as Dune. The only way it would happen is if the studios just turned it in to a generic sci-fi movie but that is something that should never ever happen, Dune is NOT Star Wars, it is NOT Star Trek it's not even Lord of the Rings. Jodorowsky's Dune is on paper at least pretty much the perfect adaption of what that world could be but it is also an impossible movie to make.

However there have been a few people who have been interested in making an animated Dune adaptation and I could totally see that working. Jodorowsky said he wanted some one to take his storyboard and make an animated movie out of it and Matt Rhodes, the guy who does that art for Dragon Age did some Dune art. Get Del Toro on board maybe get Michael Kirkbride to write it and get an animated show on Netflix.

Joss Whedon, I think would be a good director. He directed Firefly, and did one hell of a job. He's also directed a lot of other stuff like Avengers. As for actors, no clue.

Ridley Scott comes to mind for me. He is supposedly going to make a Forever War movie in the near future. It may be a good indicator if he is up to the task.
